Figure 3Differentiation capacity of mES cell–derived CPCs. mES cell–derived GFP
+ CPCs differentiate into cardiomyocytes, smooth muscle cells, and endothelial cells. Cells were dissociated and FACS sorted on differentiation day 6. Following overnight suspension/aggregation cell culture, the CPC aggregates were plated under 3 different conditions and allowed to differentiate for 7 days in culture. Differentiated CPCs stained positive for α-Actinin (Actn1;

B), Troponin-I (Tnni3;
C and
D), SMA (Acta2;
E and
F), and vWf (
G and
H; all red). Cells were also stained with phalloidin (f-Actin; green) and DAPI (nuclei; blue). Scale bars: 50μm.
